Chromatin assembly factor-1 preserves genome stability in ctf4∆ cells by promoting sister chromatid cohesion

چکیده

Chromatin assembly and the establishment of sister chromatid cohesion are intimately connected to progression DNA replication forks. Here we examined genetic interaction between heterotrimeric chromatin factor-1 (CAF-1), a central component during replication, core replisome Ctf4. We find that CAF-1 deficient cells as well affected in newly-synthesized H3-H4 histones deposition exhibit severe negative growth with ctf4∆ mutant. dissected role maintenance genome stability yeast cells. In absence CTF4, is essential for viability experiencing problems, lacking functional S-phase checkpoint or spindle checkpoint, repair pathways involving homologous recombination. present evidence affects cohesin association DNA-damage-dependent manner maintain CTF4. also show Eco1-catalyzed Smc3 acetylation reduced CAF-1. Furthermore, describe interactions genes involved loading, stabilization, indicating crucial when affected. Finally, our data indicate CAF-1-dependent pathway required functionally distinct from Rtt101-Mms1-Mms22 which functions replicated assembly. Collectively, results suggest by creates environment favors maintains integrity.

Sister chromatid cohesion and genome stability in vertebrate cells.

For successful eukaryotic mitosis, sister chromatid pairs remain linked after replication until their kinetochores have been attached to opposite spindle poles by microtubules. This linkage is broken at the metaphase-anaphase transition and the sisters separate. In budding yeast, this sister chromatid cohesion requires a multi-protein complex called cohesin. A key component of cohesin is Scc1/M...

Sister chromatid cohesion.

During S phase, not only does DNA have to be replicated, but also newly synthesized DNA molecules have to be connected with each other. This sister chromatid cohesion is essential for the biorientation of chromosomes on the mitotic or meiotic spindle, and is thus an essential prerequisite for chromosome segregation. Cohesion is mediated by cohesin complexes that are thought to embrace sister ch...

Targeted Sister Chromatid Cohesion by Sir2

The protein complex known as cohesin binds pericentric regions and other sites of eukaryotic genomes to mediate cohesion of sister chromatids. In budding yeast Saccharomyces cerevisiae, cohesin also binds silent chromatin, a repressive chromatin structure that functionally resembles heterochromatin of higher eukaryotes. We developed a protein-targeting assay to investigate the mechanistic basis...

Establishment of Sister Chromatid Cohesion

The process of sister chromatid pairing, or cohesion establishment, is coupled to DNA replication and fundamental to proper chromosome segregation and cell viability. In the past year, several articles have provided important new insights into cohesion establishment, an activity predicated on the acetyltransferase Ctf7/Eco1. Here, I review new findings that the conversion of chromatid-bound coh...
